2.The volume of the agar slant is 1/5 of the capacity of the test tube. After sterilization, it must be placed on a slanted surface while hot, and the length of the slanted surface is about 2/3 of the length of the test tube.
3.The volume of the semi-solid medium is about 1/3 of the length of the test tube. After sterilization, it will stand upright and solidify for use
4.The volume of high-layer agar is about 1/3 of the test tube. After sterilization, stand upright while it is still hot, and then solidify after cooling.
5.The liquid medium is divided into test tubes, about 1/3 of the length of the test tube.
6.Agar plate: cool the sterilized (or heated and melted) medium to about 50°C, and pour it into a sterilized plate with aseptic procedures. Pour about 13-15ml of the medium into a plate with an inner diameter of 9cm, and shake the bottom of the plate gently. , so that the medium is spread on the bottom of the dish, and it is ready after solidification. When pouring the medium, do not open the lid of the dish completely to prevent dust and bacteria from falling into the air.
The newly made plate medium (referred to as plate) has a lot of water on the surface, which is not conducive to the separation of bacteria. Usually, the plate should be placed upside down in a 37°C incubator for about 30 minutes until the plate surface is dry before use. Compared with humans, the peristaltic pump is more accurate for dispensing.
